| Abstract;Enone reductases responsible for the reduction of C=C bond adjacent to the carbonyl group of enones were isolated from cultured cells of Nicotiana tabacum, Euglena gracilis Z and Astasia longa. In order to investigate the mechanism of the enzymatic reduction, the deuterium incorporation from deuteriated medium (D2O)and co-enzyme [(4R)- and (4S)-[4-2H]NADH] to the C=C bond of the substrates was examined. It was found that the reductase isolated from N. tabacum reduces the C=C bond by syn-addition of hydrogen atoms, which is unique in the stereochemical aspect of the reduction. Using oligonucleotide primer corresponding to partial amino acid sequence of the purified enone reductase from cultured cells of N. tabacum, a cDNA fragment of 493bp containing termination codon was sequenced.
