Abstract;We have determined crystal structures of fungal nitric oxide reductase(NOR) in several oxidation states at room temperature and 77K. On the basis of the crystal structures, we prepared some mutants, examined their properties and, then, we have shown the roles of Ser286 and Thr243 in the formation of the reaction intermediate which is generated in the reaction of NOR with NO and NAD(P)H. We have also evaluated the electronic structure of the intermediate with the mixed-flow resonance Raman spectroscopy. On the basis of these results, we have proposed the molecular mechanism of the NO reduction by the fungal NOR. (author abst.)
