Abstract;The x-ray structure of catalytic antibody 7C8 has been dissoved, showint that a tyrosine in the heavy chain (H95) interacts to the oxyanion of phosphonate in the transition-state analog with hydrogen bonding. This result prompted us to examine the detailed kinetic properpies, for judging whether the tyrosine works as either transition-state stabilization or nucleophilic attack. Together with x-ray structure of 7C8, the pH profile and the hydroxylamine partitioning experiment revealed that in contrast with other antibodies within the family, the antibody 7C8 accelerates the hydrolysis with catalytic mechanism of nucleophilic attack.
