| Abstract;Conformational change of enzyme at the rate-determining step plays an important role in an acceleration of enzyme-catalyzed reactions, which can be explained by "induced-fit" hypothesis. In order to elucidate the significance of the conformational change of enzyme, we have investigated the effects of viscosity of reaction medium upon reaction rates of hydrolysis of p-nitrophenyl esters by .ALPHA.-chymotrypsin. The effects of viscosity on kinetic parameters is substrate dependent, which shows that conformational change of the enzyme depends upon the structure of a substrate. The viscosity-kinetics profile in sucrose solution is identical with that in glycerol solution, which shows that the behavior of kinetic parameters observed stems not from the specific effects of a viscogen but from the medium viscosity. The effects of medium viscosity on hydrolysis of p-nitroanilides of N-protected amino acid is under investigation. (author abst.) |
