Abstract;Acute intravenous toxicity study of amrubicin hydrochloride(SM-5887) and doxorubicin hydrochloride(DXR) at dosage of 5,7,10,12,14,16 and 18mg/kg in rats was performed, and the following results were obtained. 1) SM-5887. The LD50 values were 15mg/kg for males and 13.9mg/kg for females. Toxic signs such as decrease of spontaneous activity, ataxic gait, limb paralysis, irregular respiration, dyspnea, salivation, urinary incontinence, soft feces and/or diarrhea, piloerection, emaciation and loss of hair were observed in treated animals at 10mg/kg and above. Decreased mean body weight gains were observed at 10mg/kg and above, and suppressed mean body weight gains were observed in males at 5mg/kg and above and in females at 12mg/kg and above. Atrophy of the thymus and brown focus on the stomach for dead animals and atrophy of the thymus and hypertrophy of the spleen for schedule sacrificed animals given SM-5887 were observed by gross pathological examination. Microscopical findings caused by the suppressive effect of SM-5887 on cell growth were observed in the thymus, born marrow, spleen, digestive tract, ovary and skin. 2) DXR. The LD50 values were 11.9mg/kg for males and 14.3mg/kg for females. The same kinds of toxic signs seen in the SM-5887 treated animals were observed at 7mg/kg and above. Decreased or suppressed mean body weight gains were observed at 5mg/kg and above. Atrophy of the thymus, retention of pleural fluid, white spot of the liver and the kidney for dead animals and atrophy of the thymus, brown spot or area of the lungs, white spot, focus or white raised focus of the liver, discoloration or white change of the adrenal, atrophy or hypertrophy of the spleen, edematous change of the pancreas and retention of abdominal fluid for schedule sacrificed animals given DXR were observed by gross pathological examination....
