| Abstract;A study on intravenous administration of amrubicin hydrochloride(SM-5887) prior to and in the early stages of pregnancy was performed using rats as follows: (1) to study effects on reproductive functions of males and on development of the next generation, male rats were treated intravenously at 0.05, 0.12, and 0.3mg/kg daily during the pre-mating and mating periods, and then mated with non-treated females; and (2) to study effects on reproductive functions of females and on development of the next generation, female rats were treated intravenously at 0.015, 0.05, and 0.15mg/kg daily during the pre-mating and mating periods and in the early stages of pregnancy. 1) Effects on males. In the 0.05mg/kg group, no effects of SM-5887 were observed in general signs, body weight, body weight gain, food consumption, accumulated food consumption, necropsy findings, or organ weights. In the 0.12mg/kg group, a lower value of the testis weight was obtained. In the 0.3mg/kg group, small thymus and testes and lower values of the thymus, spleen, testis and epididymis weights were noted. For reproductive functions, no effects of SM-5887 were observed even at 0.3mg/kg in copulation or fertility. For fetuses, no effects of SM-5887 were observed even at 0.3mg/kg in the number of corpora lutea, number of implantations, implantation rate, pre-implantation loss rate, embryo-fetal death/resorption rate, number of live fetuses, sex ratio of live fetuses, body weights of live fetuses of either sex, degree of ossification, incidence of external anomalies, incidence of skeletal anomalies or variations, or incidence of visceral anomalies. 2) Effects on females. No effects of SM-5887 were observed even at 0.15mg/kg in general signs, body weight, body weight gain, food consumption, accumulated food consumption, or necropsy findings.... (author abst.) |
