| Abstract;Intravenous administration of amrubicin hydrochloride(SM-5887) at 0.05, 0.15, and 0.5mg/kg was given to rats daily during the period of organogenesis to examine its effects on dams, development and postnatal growth of the next generation, and development of the second generation. 1) Effects on dams. In the groups treated at 0.15mg/kg or less, no effects of SM-5887 were observed in general signs, body weight, body weight gain, food consumption, accumulated food consumption, necropsy findings on Day 20 of pregnancy or Day 21 post partum, or organ weights on Day 20 of pregnancy. In the 0.5mg/kg group, lower values of body weight gain in the final stages of pregnancy were obtained, and small-sized thymus and a lower value of the thymus weight on Day 20 of pregnancy were noted. For reproductive functions, no effects of SM-5887 were observed in the gestation length, gestation index, or delivery or nursing conditions. 2) Effects on fetuses(F1). In the groups treated at 0.15mg/kg or less, no effects of SM-5887 were observed in the embryofetal death/resorption rate, number of live fetuses, sex ratio of live fetuses, body weights of live fetuses of either sex, degree of ossification, incidence of external anomalies, incidence of skeletal anomalies or variations, or incidence of visceral anomalies. In the 0.5mg/kg group, lower values of body weights of live fetuses of both sexes, lower number of ossified sternebrae, higher incidence of fusion of cervical vertebral arch and higher incidence of cervical rib were obtained. 3) Effects on offspring(F1)....
