| Abstract;Amrubicin hydrochloride(SM-5887) was administered to the retroauricular vein of rabbits at 10mg/ml twice daily for 8 days to compare its vascular irritant effects to those of physiological saline solution, doxorubicin hydrochloride at 10mg/ml and 5%BSP. Administration was discontinued due to thrombus formation in 2 animals in the SM-5887 group on day 7 of administration, 2 animals each in the doxorubicin hydrochloride group on days 5 and 6 of administration and all animals in the 5%BSP group on days 3-5 of administration. Histopathological examination revealed thrombus formation, intravascular accumulation of polymorphonuclear leukocytes, edematous dissociation of the venous wall as well as hemorrhage, edema and inflammatory cell infiltration in the perivascular tissue in the SM-5887 group. In addition to similar findings, necrosis of the perivascular tissue was observed in the doxorubicin hydrochloride group. In the 5%BSP group, thrombus formation and inflammatory cell infiltration, etc. were observed, and necrosis and increased fibrous tissue were marked in the perivascular tissue. Based on these results, the irritant effects of SM-5887 noted at 10mg/ml were apparently more severe than those of physiological saline solution, slightly milder than those of doxorubicin hydrochloride at 10mg/ml and milder than those of 5%BSP.
