Abstract;We evaluated the combined analgesic effects of neurotropin and amitriptyline in SART-stressed mice which show hyperalgesia. 1. The analgesic ED50 values of single oral administrations of neurotropin and amitriptyline were 194NU/kg and 37mg/kg, respectively. 2. The combination dosages of neurotropin and amitriptyline were computed according to Gaddum's theory. In one of combinations, theoretical ED50 values(theoretical value A), [1/3ED50 neurotropin+2/3ED50 amitriptyline] was [64.7NU/kg neurotropin+24.7mg/kg amitriptyline]. In another combination, [2/3ED50 neurotropin+1/3ED50 amitriptyline] was [129.3NU/kg neurotropin+12.3mg/kg amitriptyline]. 3. Actually, We examined the combined administration with neurotropin and amitriptyline. One of two combinations of experimental ED50 values(experimental value B), [1/3ED50 neurotropin+2/3ED50 amitriptyline] was [44.8NU/kg neurotropin+17.1mg/kg amitriptyline]. Another one of combinations, [2/3ED50 neurotropin+1/3ED50 amitriptyline] was [110.3NU/kg neurotropin+10.5mg/kg amitriptyline]. 4. We calculated the ratio of theoretical value A and experimental value B, and evaluated the combined analgesic effects. In the case of [1/3ED50 neurotropin+2/3ED50 amitriptyline], B/A ratio was indicated 0.69. And in the case of [2/3ED50 neurotropin+1/3ED50 amitriptyline], B/A ratio was indicated 0.85. In both cases, B/A ratios were less than one. The combined analgesic effects of neurotropin and amitriptyline was at least more than the additional effect, because of B/A<1. (author abst.)
