| Abstract;There is no general consensus of the optimal timing of drug administration in the combined treatment of CDDP with radiation. Therefore, we irradiated the T24 human bladder cancer cell line with 7Gy X-ray and examined the effect of the administration of CDDP (10.MU.g/ml) on the cell death in three groups, comprising the simultaneous group (at the same time of drug administration), the 3-hours-later group (3 hours after irradiation), and the 15-hours-later group (15 hours after irradiation). The effect of cell death in the 15-hours-later group is stronger than the simultaneous group within 24 hours after irradiation. Flow cytometry showed G2 accumulation in the 15-hours-later group, but it did not occur in the simultaneous group within 24 hours after irradiation. The cyclin B level increasd with G2 accumulation in each of the groups. The p34cdc2 level did not increase with the cell G2 accumulation in any of the groups. In this study, CDDP should interrupt G2 accumulation and reduce the effect of cell death within 24 hours after irradiation. (author abst.) |
