| Abstract;We investigated the effects of troglitazone (Tr) on the proliferation and synthesis of transforming growth factor-.BETA. (TGF-.BETA.) and fibronectin in human mesangial cells (MC) cultured with either low or high levels of glucose. Treatment of MC with Tr caused a decrease in both DNA content and [3H]-thymidine uptake in the cells cultured with concentrations of 5mM and 33mM glucose. Incubation of MC with 33mM glucose resulted in an increase in the production of TGF-.BETA. and fibronectin compared with 5mM glucose. However, the high glucose-induced increase in TGF-.BETA. and fibronectin was halted by Tr. Accordingly, the effects of Tr on these MC functions may justify its clinical trial for treatment of diabetic nephropathy. (author abst.) |
