Abstract;We tried to determine the axial ligands of the heme in CooA that is the CO-sensing transcriptional activator. Alanine-scanning mutagenesis and EXAFS analysis have revealed that Cys75 and an oxygen atom in the peptide main chain, His77 and an oxygen atom in the peptide main chain are the axial ligands of the ferric and ferrous heme in CooA, respectively. His77 is the proximal ligand of the CO-bound CooA. The reduction and oxidation midpoint potentials of CooA were determined to be -320mV and -260mV, respectively. The fact that the reduction and oxidation midpoint potentials of CooA are different supports the proposition that the redox-controlled ligand exchange between Cys75 and His77 takes place during the change in the redox state of the heme in CooA.
