Abstract;Cocoon sericin is known to consist of several components, but their exact profiles have not yet been clarified. In this study, we dissolved cocoon silk protein in 9M-LiNCS/mercaptoethanol and separated sericin components by ethanol fractionation. Two major (-400K and 250KDa) and two minor (200K and 35KDa) sericin components were fuond by SDS-PAGE analysis of cocoon solution. The sericin components were separated by ethanol fractionation of the LiNCS solution of cocoon. SDS-PAGE of the fractions obtained showed that the components were clearly separated by this treatment. Sericin 250K component was less soluble during ethanol fractionation than high molecular weight (-400K) component. These two sericin components may differ in their chemical nature. When a cocoon was dissolved in 9M-LiNCS without mercaptoethanol, -400K and 200K components of sericin were simultaneously precipitated. This suggests these two components in a cocoon form a complex by S-S bonding. (author abst.)
