Abstract;2-Acylamino-1,8-Naphthyridine 1 that possesses hydrogen bonding groups fully complementary to guanine(G) bind selectively to a single G bulge in duplex DNA. Melting temperature(Tm) of duplexes containing a single guanine bulge increased by 5.0.DEG.C. in the presence of 1, whereas virtually no increase of Tm was observed for duplex containing adenine(A) and thymine(T) bulges as well as for fully complementary duplex. DNase I footprinting titration indicated selective binding of 1 to the G bulge with apparent association constant of 3.4.+-.1*104M-. (author abst.)
