Abstract;As a result of human autosomal translocation of t(8;21), AML 1 gene is fused to MTG 8 gene, and AML 1-MTG 8 fusion protein is expressed. Authors analyze human luekomogenesis of acute myelocytic leukemia by this fusion protein by in vitro experiments using L-G cell line established from mouse IL-3 dependent myeloid cells. AML 1 transcription factor promotes the cytodifferentiation from myeloblasts to neutrophils by forming its complex of histone acetylation enzyme including p300/CBP. AML 1-MTG 8 inhibits cytodifferentiation to neutrophils by suppressing the action of AML 1 by forming the complex of histone deacetylation enzyme mediated through MTGR 1/MTG 16. The inhibition of cytodifferentiation is found to be recovered by trichostatin A by inhibiting the action of AML 1-MTG 8.
