| Abstract;Recent knowledges obtained on the roles of AML1/PEBP2.ALPHA.B chimeric gene product in leukomogenesis are explained by two approaches of loss-of-function and gain-of-function of AML1. AML1 is essential for cytodifferentiation of bone marrow cells, and its loss-of-function causes increase of leukemia cells with abnormality of proliferation by suppressing normal differentiation of stem cells. As the examples of loss-of-function of AML1, authors discovered leukemia with point mutations in AML1,and hereditary disease of familial platelet disorder with predisposition to acute myelogenous leukemia with mutation in AML1. And the mechanisms of AML1-related leukomogenesis in humans are discussed. |
