| Abstract;We studied in vitro glucose absorption from the small intestine via isolated perfusion in GK rats, an animal model of type 2 diabetes mellitus. GK and Wistar rats 10 to 13 weeks old were made to fast for 24 hr and perfusion conducted as described by Levin et al. The superior mesenteric artery was perfused with KRB containing 60, 140, or 300 mg/dl glucose. Luminal perfusion was started with 3% glucose solution, 20 min following vascular perfusion, for 30 min. Glucose absorption was estimated from the increase in glucose concentration in portal venous effluent. The integrated increase of glucose concentration in portal venous effluent was sigufficantly decreased in Wistar rats, when glucose concentration in the arterial perfusate was increased to 140 and 300 mg/dl. No change was found, however, in GK rats. The glucose concentration in portal venous effluent from the small intestine was greater in GK rats than in Wistar rats, suggesting that elevated blood glucose in spontaneously diabetic GK rats are, at least partially, attributable to increased glucose absorption from the small intestine.
