Abstract;The effects of galanin and its non-selective receptor antagonist (galanin-A) [D-Thr6,D-Trp8,9,15-ol]-galanin1-15 on cell proliferation in immature rat adrenal glands have been investigated by metaphase-arrest and PCNA-immunostaining techniques. Galanin-A alone or with galanin lowered PCNA index in the zona glomerulosa (ZG), without affecting it in zona fasciculata-reticularis (ZF/R) and adrenal medulla (AM). In contrast, galanin-A increased metaphase index in AM, the effect being potentiated by the simultaneous administration of galanin. Metaphase index in ZG and ZF/R was not affected. Galanin per se did not change metaphase or PCNA indexes in immature rat adrenals. The conclusion is drawn that endogenous galanin exerts a tonic control of adrenal growth in immature rat: it stimulates the entry of ZG cells in the S phase and lowers the progression of AM cells in the M phase. Our investigation also indicates that the combined use of metaphase-arrest and PCNA-immunostaining methods is essential to investigate factors modulating adrenal-cell proliferation. (author abst.)
