| Abstract;In this study, we investigated the expression pattern of versican in oral epithelia both in vivo and in vitro. Porcine periodontal tissues and epithelial rests of Malassez derived from periodontal ligament were employed. Immunohistochemical analysis was carried out using versican antibody (5D5) and cytokeratin 19 antibody (CK19). The protein and mRNA expressions were confirmed by western blot and RT-PCR, respectively. In the oral epithelium, the basal and spinous intercellular spaces showed intense staining for 5D5. In the junctional epithelium, intense staining was observed in one or two cell layers near the enamel surface. Epithelial rests of Malassez in a high Ca2+ condition expressed higher levels of versican than those in a low Ca2+ condition. The localization of versican in the epithelial cells was slightly more differentiated than cytokeratin 19. These results suggest that versican plays an important role in the physiological maintenance of oral epithelia. (author abst.) |
