| Abstract;The active control of the DNA/RNA recognition by an external/internal factor should be essential for the next generation of antisense molecules. If one could control the syn-anti orientation of switching of a nucleobase through the application of an external factor or agent, active control of the recognition and complexation behavior of antisense molecules could be achieved. For this purpose, a novel nucleic acid model using peptide ribonucleic acid(PRNA), which contains 5'-amino-5'-deoxypyrimidine ribonucleoside as a recognition site for nucleic acids, has been designed, synthe-sized and applied to the external reversible control of recognition behavior of the complementary polynucleotide through the orientational switching of the nucleobase induced by borates. In this paper, in order to extend this methodology, the effects of added borates on the nucleobase orientation and recognition behavior of novel mono-, oligo-, and polymeric PRNAs containing 5'-amino-5'-deoxypyrimidine and purinenucleosides were investigated. In case of PRNA oligomer containing pyrimidine-purine mixed sequence, efficient orientational change of nucleobases was observed. (author abst.) |
