Abstract;In vitro selection has enabled us to design molecular recognition DNA that is called aptamer or to design catalyic DNA or RNA that is called DNAzyme or ribozyme, respectively. In the present study, the molecular recognition DNA and DNAzyme were combined to be a DNA nano-device, which can be used as antibody-enzyme conjugate used for Enzyme Immuno Assay(EIA). Peroxidase activities of various DNA molecules, which were reported to interact with porphyrin, were investigated. Among the DNAs the highest active DNA was conjugated with thyroxine-binding DNA molecule, which was previously reported by us, through spacer DNA sequence. The designed DNA was synthesized and was employed in the EIA experiments. (author abst.)
