Abstract;Background Skin fibroblasts are mesenchymal cells ubiquitously present in dermis, produce structural proteins of connective tissues such as collagen and play important roles in wound repair and others. Recently, it was found that IL-4 stimulated eotaxin production in fibroblasts. As eotaxin induces chemotaxis of eosinophils, the role of fibroblasts as immunomodulatory cells in the pathogenesis and progression of allergic skin diseases, such as atopic dermatitis, is getting attention. Objective We examined the effects of histamine on the production of eotaxin in skin fibroblasts and the inhibitory effects of epinastine, an anti-allergic drug. Methods We cultured normal human fibroblasts, replaced the medium with serum-free medium when they were semiconfluent and stimulated them with histamine. Forty Eight hours later, we collected the medium and the cells and measured the production of eotaxin proteins by ELISA and the expression of mRNA by RT-PCR. we examined effects of epinastine and by adding them to histamine or IL-4. Results Histamine increased the production of eotaxin in the fibroblasts in dose-dependent manner. Epinastine inhibited histamine-induced production of eotaxin protein and the expression of mRNA but it did not inhibit the massive production of eotaxin induced by IL-4. These results indicate that epinastine might inhibit eosinotaxis stimulated by histamine-induced eotaxin and has clinical effects in allergic diseases. (author abst.)
