Abstract;This study was conducted to determine if IL-8 activates EGF receptor on gastrointestinal epithelial cells via membrane anchored metalloproteinase, ADAM, which releases EGF ligands from membrane anchored precursors. IL-8 phosphorylated EGF receptor on Kato III cells within 15min. This phosphorylation was significantly blocked by pretreatment with ADAM inhibitor 7795. These data indicate that IL-8 may stimulate a growth of gastrointestinal epithelium by transactivating EGF receptor in an ADAM dependent manner. This system including ADAMs and EGF receptor ligands may play a key role in various pathological condition of gastrointestinal disorders. (author abst.)
