Abstract;Lithium is a mood stabilizer used in treatment of bipolar disorders. We reported that GAP-43 protein was induced after 4-5 weeks lithium treatment. We examined the GAP-43 RNA expression after longterm treatment with lithium in the rat brain. GAP-43 RNA also increased in rat cerebral cortex after 4-5 weeks treatment with lithium. The E-box binding protein to the GAP-43 promoter was decreased after 4-5 weeks lithium treatment. Since the decrease of E-box binding protein preceded increased GAP-43 mRNA expression, it is suggested that E-box binding protein to the GAP-43 promoter could negatively regulate GAP-43 gene expression. It was thus suggested that the lithium increase in GAP-43 mRNA and GAP-43 protein level by E-box binding protein. GAP-43 is known to play an important role in the releasing process for several neurotransmitters(Ach,5-HT and DA). It was considered that the long-term lithium treatment regulated GAP-43 and release of neurotransmitters by E-box binding protein. (author abst.)
