| Abstract;Laminin, which is one of the extracellular matrix components and distributes around senile plaque in Alzheimer's disease, has a role to inhibit the aggregation of beta-amyloid peptide(A.BETA.) to suppress the neurotoxicity of A.BETA.. Recent investigations have explored A.BETA.-induced neurotoxicity would be derived from free radical-induced lipid peroxidation. The aim in this study is to confirm whether laminin has an antioxidant effect to play a neuroprotective role on the neurodegeneration. To confirm it, radical scavenging ability of laminin, fibronectin and bovine serum albumin on the generation of hydroxyl radical in the medium of Fenton reaction or H2O2 irradiated with 345nm UV was investigated. Although there was no specificity in the inhibition of Fenton reaction induced-hydroxyl radical generation among these materials investigated here, hydroxyl radical scavenging ability of laminin was significantly higher than that of the others. From the results, laminin has potential to inhibit growing reaction of lipid peroxidation following hydrogen abstraction by hydroxyl radical. It will be possible to conclude laminin, which is one of the extracellular matrix components to inhibit A.BETA. aggregation has the effect of an intracerebral antioxidant on lipid peroxidation.
