Abstract;SMS-113 is a novel over-the-counter analgesic drug containing ibuprofen, tranexamic acid and caffeine. In this study, the anti-inflammatory and analgesic effects of SMS-113 in rat carrageenin-induced paw edema and the Randall-Selitto test were compared with those of reference analgesic drugs. The anti-inflammatory and analgesic effects of SMS-113 (1/10 of oral dose of adult human/kg rat) were almost equal to those of other ibuprofen-caffeine combination analgesic drugs, and stronger than analgesic drugs featuring combination with aspirin or acetaminophen. However, SMS-113 tended to have quick and long-lasting effects. Additionally, we evaluated the gastric irritation in rats and prolonging of pentobarbital-sleeping time in mice of SMS-113 and other reference analgesic drugs. The gastric injury by SMS-113 (two oral doses of adult human/kg rat) was weaker than that by other reference analgesic drugs in rats. Furthermore, SMS-113 (one oral dose of adult human/kg mouse) did not prolong pentobarbital-induced sleeping time in mice. These results suggest that SMS-113 is a useful over-the-counter analgesic drug with quick and long-lasting effects, and without the side effects of gastric injury and sleepiness. (author abst.)
