Abstract;High-density lipoprotein(HDL) plays a principal role in reverse cholesterol transport(RCT) from peripheral tissues to the liver. RCT consists of 3 steps;(1) removal of cellular cholesterol by lipid-poor apoA-I and generation of lipid-poor HDL(=pre .BETA.1-HDL),(2) maturation of lipid-poor HDL into spherical HDL by lecithin-cholesterol acyltransferase, and (3) uptake of chloesterol of spherical HDL by the liver. This review describes the mechanisms of these steps, and summarizes the recent advances in the research on ATP-binding cassette transporter 1(ABCA1), the critical membrane-associated protein in the step 1 of RCT.
