| Abstract;F1 intergeneric hybrid of 2n=27(triploid) was artificially produced between the maternal Dendranthema horaimontana(2n=18) and the paternal Tanacetum parthenium(2n=36). The parental genome identification, organization and relationship were analyzed in somatic nuclei of root-tip-cells of the sexually mature F1 hybrid plants. The two genomes were identified in spread material by non-radioactive DNA: DNA genomic in situ hybridization using biotin labelled total genomic DNA from one of the parents as a probe. The genome territories of D. horaimontana displayed as nine separated yellow-colored chromatin domains or nine chromosome territories in the interphase nuclei after hybridization with the labelled total genomic DNA of D. horaimontana probe. In contrast, the genome territories of T. parthenium could not be identified as 18 separated-chromatin domains. The organization of the nine-chromosome territories of D. horaimontana were not oriented according to the Rabl model in interphase nuclei. At metaphase spread the two parental chromosomes have been distinguished as a nine yellow-colored chromosomes of D. horaimontana hybridized with the labelled total genomic DNA of D. horaimontana probe and 18 red-colored chromosomes of T. parthenium did not.
