| Abstract;From results of large landmark clinical randomized controlled trial, such as 4S, WOS, CARE, LIPID, AFCAPS/TEXCAPS, and HPS, inhibitors of HMG-CoA reductase (statins) have shown significant reductions in the incidence of cardiovascular disease and stroke. Beyond lipid alteration, statins have pleiotropic effects not only on vascular but also on extravascular system including bone, brain, retina and carcinogenesis. These effects of statins may be related to the down-regulated intermediate substances produced from mevalonate pathway, as prenylated proteins, dolicol, ubiquinone. In future, statin may be indicated for additional therapy to osteoporosis, dementia, Alzheimer disease, hypertension, macular degeneration and even to cancer. (author abst.) |
