Abstract;Hepatitis C virus is a major cause of chronic hepatitis that progresses to liver cirrhosis and hepatocellular carcinoma. Only available therapy for chronic hepatitis C is the treatment based on IFN. However, genotype 1b, that is a most prevalent genotype in Japan, is interferon-resistant strain. We investigated the relation of HCV genetic structure to the response to interferon, and found the interferon sensitivity determining region (ISDR: aa.2209-2248) in NS5A of genotype 1b. NS5A reportedly inhibits the interferon induced double-stranded RNA-dependent protein kinase (PKR) which plays a key role in the antiviral and antiproliferative effects of interferon. Recent development of HCV replicon system enables us to investigate the effect of IFN/Ribavirin against HCV in vitro. (author abst.)
