Abstract;Effect of a novel AT1 receptor antagonist telmisartan on human platelet function was investigated. P-selectin expression and PAC-1 binding as markers of platelet alpha-granule secretion and activated glycoprotein (GP) IIb/IIIa, respectively, were analysed in platelets exposed to 300nM of thromboxane A2 (TXA2) analog U46619 by flow cytometry. Telmisartan significantly inhibited the P-selectin expression and GP IIb/IIIa activation at a concentration of 10.MU.M. Platelet aggregation was measured with an aggregometer, initiating the aggregation reaction either by the addition of U46619 or collagen. Telmisartan significantly inhibited both of stimulant-induced platelet aggregation at a concentration of 50.MU.M. These results indicate that telmisartan inhibits platelet activation and aggregation induced by TXA2. (author abst.)
