Abstract;Preventive effects of telmisartan, a novel angiotensin AT1 receptor antagonist, on the neointimal hyperplasia and the endothelial dysfunction after balloon injury were examined in rat carotid artery. Two-week repeated oral administration of telmisartan at 10mg/kg/day significantly decreased both systolic and diastolic blood pressure. Telmisartan improved intimal thickening (I/M ratio) by 54%. Acetylcholine-induced vasodilation was reduced in injured arteries (maximal relaxation; 64.+-.6% in injured versus 86.+-.2% in normal, P<0.01). Telmisartan was significantly ameliorated the impairment of vascular relaxation (maximal relaxatiorn; 90.+-.5%, P<0.01). However, there was no difference in sodium nitroprusside-induced vasodilation among all groups. Telmisartan also inhibited angiotensin II-induced contractile response (maximal contraction; 62.+-.9% in injured versus 17.+-.5% in telmisartan, P<0.01). These results suggest that telmisartan prevents intimal formation and atherogenic impaired endothelial vasodilation following endothelial injury. (author abst.)
