| Abstract;This study was conducted to investigate the effects of differentiation stage of donor cells on developmental capacity of bovine nuclear transfer embryos reconstructed with cranial neurocyte. First, we examined the culture method of bovine cranial neurocyte for using donor cells in nuclear transfer. Bovine cranial neurocytes derived from fetuses from slaughterhouse were successfully cultured in Neurobasal medium in poly-L-lysine coated plastic dishes at 39.DEG.C. in 5% CO2 in air. Next, the cultured neurocytes and bovine amniotic cells(control) were used for donor cells in bovine nuclear transfer. The frequencies of fusion, cleavage and development to the blastocyst stage of the reconstructed embryos derived from neurocytes were lower significantly(P<0.05). No blastocyst was obtained from the reconstructed embryos derived from neurocytes. These results suggest that there may be another optimal condition of the nuclear transfer using neurocytes and/or the reconstructed embryos derived from neurocytes may have low capacity for development.
