| Abstract;In order to observe the early events of protein folding, we developed a new method, modification of a specific amino acid with a photocleavable group and subsequent irradiation with ultra violet light to the modified protein. Plastocyanin (PC), which is an electron-transfer protein containing beta sheets, unfolded partially by the modification. Subsequent light irradiation to the modified protein initiated cleavage of the modified group and led the protein to refold. We determined the modified amino acid and elucidated that PC refolds from the partially unfolded structure to the native structure by cleavage of the modified group with the irradiation. The fast refolding reactions of PC are analyzed with the transient grating method. (author abst.) |
