| Abstract;In order to help understand growing mechanism of smooth muscle cells in the vascular modeling, in vitro experiments were performed by flow cytometry, Western blotting, gene introduction, etc. using cultured rat thoracic aortic smooth muscle cells. As the result, when the serum was applied to the smooth muscle cells, the progress in the cell cycle was promoted, the expression of various cell cycle markers was increased and the activity of the cyclin-dependent kinasewas also increased, but the smooth muscle cells were not expanded. When angiotensin II was applied to the smooth mucle cells, the cell diameter was increased significantly. The p21Waf1 expression in the smooth muscle cell was increased when anigiotensin II was applied, but was not increased when the serum was applied. When p21Waf1 gene was introduced, p21Waf1 was overexpressed, resulting in not only a decrease of cell in S phase but also cell hypertrophy. The antisense oligodeoxynucleotide for p21Waf1 inhibited the cell hypertrophy by angiotensin II. From the above, there seemed to be a possibility that p21Waf1 fulfill some roles in the hypertrophy of vascular smooth muscle cells. |
