Abstract;The following were examined : 2 kinds of single base polymorphism of endothelin (ET) 1 gene (EDN1), Lys 198Asn (5727G>T) and 3A/4A (-134delA). As a result of manifesting Lys type or prepro ET1 of the Asn type in COS1 cells, 293 cells, human umbilical vein endothelial cells (HUVEC), it was indicated that there was no significant difference in the concentration of ET1 and big ET1 in the culture supernatant, and that the Lys 198Asn polymorphism did not affect the ET1 production. As a result of examining the difference in ET1 concentration in the blood plasma between genotypes in 54 cases of essential hypertension patients, the patients with Asn allelomorph tended to be higher in ET1 concentration in the blood plasma than 198Lys homozygote, but there was no significant difference. The significant difference was recognized in ET1 concentration in the blood plasma between 3A/3A and 3A/4A.
