| Abstract;Fosfomycin (FOM) is an antibiotic with a unique structure, and known to have an excellent safety profile. In the present study, we investigated the cross-reactivity of FOM with widely used .BETA.-lactam antibiotics such as ampicillin (ABPC), cephalothin (CET), ceftizoxime (CZX), cefazolin (CEZ), cefotiam (CTM), imipenem (IPM), and aztreonam (AZT) in delayed type hypersensitivity reaction in guinea pigs as part of safety assessment. As a result, the animals sensitized with FOM showed no hypersensitivity at the FOM challenge or cross-reaction at the challenge with the other antibiotics tested. On the other hand, hypersensitivity was observed in the animals sensitized with CET at CET or CTM challenge, and the animals sensitized with IPM at the IPM challenge. These results indicate that FOM has minimum antigenicity and cross-reaction against .BETA.-lactam antibiotics, which confirmed its safety in clinical use. (author abst.) |
