| Abstract;In this research, we focused on the association between highly malignant lung cancer with neuroendocrine differentiation and proliferation and examined its significance in the onset and development of cancer. We performed examinations to develop a new lung cancer treatment using RNAi. As a result of forced expression of master gene ASCL1 for neuroendocrine differentiation, we observed the induction of neuroendocrine differentiation markers and expression suppression of factors that negatively regulates cell cycles. We suspected that ASCL1 accelerate the dell cycles through transcription suppression. Moreover, we demonstrated that suppression of ASCL1 by RNAi induces cell cycle suspension and causes inhibitory effects for cell proliferation which is remarkable specifically in lung cancer cells that express ASCL1. The cancer gene therapy using RNAi may become a safe, cancer-specific therapy. |
