Abstract;Excessive host immune responses against the infected or target-loaded hepatocytes may be the key mechanism of massive livernecrosis in the fulminant hepatitis. And there are accumulating evidences that cytokines are involved and play important roles in this mechanism as the inflammatory mediators. Both in the hepatitis model of hepatitis B virus-transgenic mouse and in the hepatitis model of mouse or rat administerd with Propionibacterium/LPS, TNF-.ALPHA. and IFN-.GAMMA. are suggested to be key cytokines that cause severe liver injury. Furthermore, it is reported that the production or the expression levels of cytokines are elevated both in the serum and in the liver of the patients with fulminant hepatitis. Thus anti-cytokine therapy, such as anti-TNF-.ALPHA. and anti-IFN-.GAMMA. therapy may be the promising candidates for the treatment of fulminant hepatitis and need to be investigated further.
