Abstract;Characterization of T 2 and 6 C 4 mAbs by western blot. Blot signals are representative of 100 mg of tissue of either mouse scrapie, sheep scrapie, or CJD. The detected abnormal PrP'res' bands (known to derive from Prp33-37 residues through removal by PK of 67 amino-terminal residues) are evidenced as small bands of PrP27-33 (Bolton et al, 1982). MAbs T 2 and 6 C 4 reacted clearly with PrP'res' mouse and PrP'res'-sheep at a dilution of 1 : 5000 (Fig.1). PK completely digested PrP'c' of uninfected mouse and sheep tissues. Consequently, no reactivity was obtained (data not shown). Strong signals were observed for PrP'CJD' with T 2 and 6 C 4 (Fig.2). The two mAbs recognized both PrP'BSE' and PrP'sc'-sheep of Japanese strain very well. Again, human PrP'c' did not show any reactivity with these mAbs (data not shown). Detection of PrP'res' in paraffin-embedded sections. Kuru-type spherical plaques present in cerebellum of a CJD case with codon 129Met/Val genotype were stained after formic acid pre-treatment (van Keulen et al, 1995; Bell et al, 1997) using T 2 (1 : 2,000) and 6 C 4 (1 : 5,000). The staining pattern of plaques was marginal for T 2 and central for 6 C 4 (Fig.3). Positive plaques were revealed predominantly in the granular layre with occasional plaques in the molecular layer of type- 2 CJD. Synaptic type (type-1) PrP'CJD' was also revealed by both mAbs (T 2 and 6 C 4). Diffuse fine deposition around the remaining neurons in the affected cortex was seen. In the BSE affected brain, diffuse particulate PrP staining of neuropile was seen with T 2 at a dilution of 1 : 2,560 and with 6 C 4 at a dilution of 1 : 10,240 (Fig.). No reaction was detected in tissues from uninfected humans or cows. (author abst.)
