Abstract;We have recently identified a 17-kb chromosomal region which contains a type III secretion system gene cluster in Enterohemorrhagic Escherichia coli (EHEC) O157, and designated E. coli type III secretion 2 (ETT2). To determine the degree of ETT2 distribution among O157 isolates, and the possible involvement in the O157 pathogenicity, we examined the presence of the ETT2 in two hundred two O157 strains (73 human and 139 cattle isolates) and 12non-O157 strains (2 human and 10 cattle isolates) by PCR-based analysis for some virulent genes and three locus (L, D, and C) of the ETT2, and compared about their genetic profiles. By such broader surveys, the ETT2 gene cluster was present in the majority of O157 strains isolated from human and cattle, especially in Shiga toxin-producing E. coli (STEC), but not in common among non-O157 strains. Southern blot hybridization, and sequence analysis on ETT2C lous revealed that an one-base difference was found between human and cattle isolates in the sequence of a certain gene. The ETT2 locus might participate in the pathogenicity of theses bacteria unknown manner. However, it remains under whether this difference influences the biological function of the ETT2 gene cluster. To evaluate the ETT2 as a good marker for molecular epidemiological surveys of O157 strains, further investigation should be required. (author abst.)
