Abstract;A simple and reliable method using liquid chromatography-electrospray ionization-mass spectrometry (LC/ESI-MS) has been developed for the determination of chloramphenicol in meat and fish. The drug was extracted from meat and fish with 0.2% metaphosphoric acid-methanol (6 : 4), and the extracts were cleaned up on an Oasis HLB cartridge (60mg). The negative ionization produced the molecular related ions, (M-H)'-', at m/z 321 and 323 for chloramphenicol. The LC separation was performed on a Mightysil RP-18 column (150 * 2mm i.d.) using 10mM ammonium formare-acetonitrile (75 : 25) as the mobile phase at a flow rate of 0.2ml/min. The molecular related ions of the drugs observed under the established conditions were excellent. The calibration graph for the drug was rectilinear from 0.005 to 1.0.MU.g/ml with selected ion monitoring (SIM). The recoveries of the drug from meat and fish fortified at a level 0.05.MU.g/g was 73.2-90.2%, with high precision. The limit of detection of the drug in meat and fish were 0.001.MU.g/g. The developed analytical method might be applicable to the estimation of the residual carcinogenic antibacterials in livestock products. (author abst.)
