| Abstract;The effect of two kinds of 1,4-dihydropridine calcium-channel blockers, nicardipine hydrochloride and nifedipine, on the disposition of carvedilol, was studied in rats. The plasma carvedilol concentration was found to be significantly higher, and the AUC was higher after simultaneous oral administration of carvedilol with nicardipine hydrochloride or nifedipine, respectively, than after administration of carvedilol alone. The plasma carvedilol concentrations were significantly higher than the controls and the drug pharmacokinetic parameters Tmax was shorter, while Ka and AUC were higher after the oral administration of metoclopamide. In contrast, propanthine bromide, which delays gastric emptying, slowed absorption, but not according the AUC of carvedilol. The administration of carvedilol with colestimide did not change the dependent profiles of the plasma concentration or pharmacokinetic parameters of carvedilol. (author abst.) |
