| Abstract;The hair follicle bulge area is an abundant, easily accessible source of actively growing, pluripotent adult stem cells. Nestin, a neural stem cell marker protein, is also expressed in hair follicle stem cells as well as their immediate differentiated progeny. Nestin-expressing hair follicle stem cells give rise to the outer-root sheath and the interfollicular vascular network. The nestin-expressing hair follicle stem cells are positive for CD34 but negative for keratinocyte marker, keratin 15, suggesting their relatively undifferentiated state. Hair follicle stem cells can differentiate into neurons, glial cells, keratinocytes, smooth muscle cells and melanocytes in vitro. Neural-crest-like stem cells have also been identified in the bulge area from mouse whisker hair follicles, and neural-crest-like stem cells differentiate into neurons, smooth muscle cells, glial cells, and melanocytes in vitro. Nestin-expressing hair follicle stem cells promote the recovery of peripheral nerve injury. Hair follicle stem cells derived from a .BETA.-actin-driven-GFP transgenic mouse, implanted into the gap region of a severed sciatic nerve in a C57BL/6 immunocompetent mouse, greatly enhance the rate of nerve regeneration and the restoration of nerve function. Hair follicle cells transdifferentiate largely into Schwann cells, which are known to support neuron regrowth. The rejoined sciatic nerve contracted the gastrocnemius muscle upon electrical stimulation. After severing the tibial nerve and subsequent transplantation of hair follicle stem cells, walking print length and intermediate toe spread significantly recovered. Another study demonstrated that neural-crest-like stem cells in the hair follicle bulge area differentiate into GABAergic neurons, and intermingle with host neurites in the injured spinal cord....
